Your can separate and measured Indigo and Leuco indigo dyes based on any reversed phased HPLC. But you must be optimized all of the experimental conditions.

I found that a recent bachelor thesis (2019) may be suitable because a lot of experimental details are given. Please see attached document.
At first, check the separation methods related to it. Then identify the desired compounds using the available standards. Also, the relevant protocols could be easily searched in online and hard copy sources.

The method must exist, but It will be less expensive (time and coast) with spectrophotometer UV-Vis.
We need to distinguish between Indigo and leuco indigo. During reaction monitoring we need to identify unreacted indigo in Leuco indigo (liquid indigo). Thats why we are working on HPLC method.
