I am trying to preserve the anaerobic granules for Hg Porosimetry test. I can fix the granular sludge using 2.5 % Glutaraldehyde solution but the test requires dried sample. Some researchers have used oven drying but it contracts the shape of granule and Some have used graded ethanol solution to dehydrate.
Does anyone have experience using any of these or other protocol. ?

Hg porosimetry indeed requires dry samples. You can freeze dry your sample, and check that its volume did not change (no shrinkage). However, there is a risk of changes of pore size changes, due to the crystallisation of the solvent used for freeze drying. Supercritical drying in CO2 therefore will avoid this problem, but is not available to everyone.
