For DNA isolation from Gram positives whatever kit you use it should have enzyme Lysozyme separately for the thick cell walls of gram positive bacteria and yeast so follow the protocol as per kit, but the most important aspect that this lysozyme usually alone is not enough (as I myself have seen it several times) to do the best job of cell wall lysis so any how you need to perform mechanical homogenization or disruption on standard overnight grown bacterial inoculum in the lysis buffer as mentioned in your DNA extraction kit. Usually, 0.1 mm glass beads are fine for such bacteria so perform it in some homogenizer at good speed then centrifuge well, take the clear supernatant and perform routine extraction protocol of your kit column based or else..

Thanks Yousuf Ali Sarker vaiya, I tried this method with some of modification for direct dna extraction from soil sample. This protocol you showed is for culture bacteria. But We want directly from soil. However, Thank you very much. It will very much helpful I hope when we can culture our bacteria.

Oh sorry Vaiya, I think I couldn't explain my word. I found a protocol from a paper that only work for soil sample, not bac colony. The protocol you gave is similar to that of my protocol but not almost. But thanks for this, because It will help me for another study. :-) 

I have used the Mobio PowerMax soil extraction kit for extraction of genomic DNA from sediment samples that contained humic substances with good results (see the link). I was able to extract ~1 ug of DNA per gram of soil, however concentration of the DNA was required to get a concentration required for sequencing library prep.

In the standard DNA extraction protocol that I mainly use for isolate cultured but it can be used on environmental samples, a enzymatic lysis step which includes lysozyme and achromopeptidase (from Achromobacter, sold by sigma) breaks down the cell wall of gram positive bacteria allowing SDS to penetrate and burst the cell membrane.

Hello Jawwad Ahmad , I was reading your answer for DNA extraction of Bacillus strains. I am extracting the DNA of lactic acid bacteria (Gram +) which are being difficult to extract by using a commercial kit (UltraClean Microbial DNA Isolation Kit, MOBIO Labs) which is based in a combination of heat, detergent and mechanical forces (microbeads). As recommended by the manufacture, we also tried to lysate the cells by heating up at 70 °C. It helped a bit for some strains but not for others.

My question is how we can include the enzymatic digestion step with lysozyme in the commercial isolation method. Do you suggest a reference protocol for this?
