Maybe using another adhesive slide or coating your slides with L-polylysine could solve your issues. I used superfrost plus slides in the past and they worked well or my samples. There are also Star frost slides (knittel glass), which are meant to be good as well. Hope this helps.

I don't know what kind of slides you use, but the superfrost slides are supposed to have coating that helps with adhering the tissue. I also let my tissue dry onto the slide before starting my staining. Another difference is I use shandon M-1 embedding matrix instead of OCT because I find it's much easier to cut with, although that may not affect your tissue detachment problem, but I do get fewer wrinkles which gives less chance for the tissue to detach. And I also rock my slides pretty gently during staining.

Dear Gustavo, Osteochondral tissue is extremely difficult to keep on slides, avoid wrinkles etc. I have worked a long time on rheumatoid and osteorthritic cartilage (human) some frozen, but we moved to paraffin wax embedding eventually. I have used polylysine and APES (from Sigma) coated slides and APES coating gave me slightly better adhesion of my sections. I work now on different types of frozen samples with OCT and Cryomatrix - the latter is very good. I use Surgipath X-tra Adhesive slides (Leica Biosystems - check on their website). Good luck with your work. 

Agree with Maria, move to paraffin embedding. You still can embed into paraffin even already embedded in OCT. But paraffin bone are still fall off if you didn't fix and decal well, especially when you do antigen retrieval.  But you don't want over fix and over decal, otherwise you lost antigenicity.

I work in an orthopedic research lab and highly recommend the Kawamoto tape transfer technique.  Although the supplies can only be purchased from Section-Lab in Japan (http://section-lab.jp/), the resulting quality of section, as as well as tissue retention, is amazing!

Agree with Carol. Kawamoto's method is the best so far for frozen undecal bone sectioning. 

Gustavo - I assume you are working with frozen tissue since you embedded in OCT...were they run through gradiated sucrose after fixation? It sounds like some here are not familiar with the CryoJane system. You have to use their slides. You can try using the 4x adhesive slides which have more adhesive for use with bone and fatty tissue than the 1x slides. Also double flash with the UV. Be sure the tape is well adhered to the tissue usign the roller before sectioning. Section thin (3-5 um). When peeling the tape from the slide start at on corner and peel back slowly and diagonally at a sharp angle. You are working with a tough tissue for frozens. Consider decalcification via EDTA chelation which is gentler on the tissue then acids, takes longer, but preseves much antigenicity. Be very gentle with all the steps during the IHC process, particualrly the washes. Last, as already mentioned, since these are fixed maybe processing and paraffin embedding will alleviate some of the pain.  Good luck..

I also faced same problem once and than changesd the brand  and used the silanized one, also i faced same problem when we were using self prepared Poly-lysine coated slides.Better select Good Brands or try silanized Slides

Thanks for the useful hints. Does anybody know how to order Kawamoto tape transfer kit and stuff from Japan? Their website is really old fashioned and hard to find information. I need to do IF on mouse long bones for stem cells.

I face the similar kind of problem.

I use Lysin coated slide for wax embedded tissue. Though my section do not fall off but the edge of the sections fold that destroys my whole day work.
