I am doing a project on Matrix Tablets using natural products as binders (release retardant polymer) and I came across Swelling Behavior/Index. Is this necessary? I know swelling is vital for gel formation and slow release of drug from matrix. Is there a limit or acceptance criteria?

In sustained release I think the main factor to be considered is the release rate of the drug you include in your matrix. To achieve that release rate, you may use the combination of polymers and other excipients that you can. So I don not think there is a limit for the swelling index behavior.
I think not. But you may control by varying polymer amount it according to your desired outcome. Swelling may be a factor in releasing drug. By maintaining it, you may get your desired result. DoE may be applied to fix it. By using RSM , you may get an optimize value.
