I've been unsuccessful in all attempts of a wet mount preparation of a 5-part WBC differential using various Romanowsky stains (e.g. Wrigth-Geimsa, Wrights, Geimsa). The protocol has entailed various ratios of stain to whole blood / dilution of whole blood and incubating between 5 and 10 minutes.

I've found that the cells are not fully stained and do not show a difference in cytoplasm, nucleus, and granules.

I am not looking for a blood smear protocol. This scenario requires a wet mount protocol only.

Would you have any suggestions on stain and protocol?
