Basically i want to inquire if we treat gelatin protein solution with liquid nitrogen for prolonged time (1hr, 2hr, 3hr, 4hr...etc) and fabricate gelatin film using freeze drying. Now, can we expect any difference in tensile strength between cryo-treated gelatin film and without treated gelatin film.
By freeze drying you might be able to obtain a gelatin dry film of relatively high porosity. Of course, alternative drying techniques would result in gelatin films (or powders) having different characteristics. Of significant importance to determine the characteristics of the film would be the concentration of the prior gelatin solution. After cooling, you would obtain a frozen hydrogel with ice crystals dispersed within the gel network. I would say that, being the only parameter changed, the time during which the frozen gel is kept at a constant low temperature is not expected to significantly alter the characteristics of a gelatin film obtained by freeze drying. Also to be noticed is that, while being freeze dried by current technology, the previously frozen gel would have meanwhile been thawed to a higher temperature than that previously used to keep the gel frozen for the selected period of time.
if i keep the concentration same for a) Untreated gelatin solution and b) Treated gelatin solution (cryo treated for 5 hr) can i expect any alteration in secondary structure (a-helix, b-sheet) in Circular Dichromism(CD) of both the gelatin sample?
I think that the freeze-thawed gelatin solution may possibly show some kind of noticeable difference when compared with a ‘control’ solution (not freeze-thawed). Such a distinction may, in principle, occur at the level of the colloidal interaction between the gelatin molecules and water dipoles, since colloidal solutions or sols may be able to evidence hysteresis related effects, particularly by temperatures close to the sol―gel transition temperature or the sol―gel temperature transition range. Circular dichroism (CD) measurements seem not particularly suitable to investigate such non-linear time-dependent effects. In principle, I would not expect any significant change at the level of the molecular gelatin structure due to the freeze thawing thermal history. Dynamic light scattering (DLS) would be better suited to investigate such (seemingly possible) time-dependent effects of the colloidal solutions.
