There is controversy in literature, where some report not to use more then 2.5% DMSO and few report 100% DMSO can be used to dissolve your extract to testify its antimicrobial activity. Kindly suggest whether to use 100% DMSO or any other solvent. Even some recommend ethyl acetate, which is non toxic.

All solvent must be having some adverse effect on the microbial growth. However, DMSO is generally used to dissolve and assay for antibiotic compounds. You may preferably use 100% DMSO and you may also maintain a DMSO control. That would serve the purpose

DMSO has been used at varied concentrations as you have mentioned. For antimicrobial activity 100% may not be a problem. For cytotoxicity and cell line related assays you may consider lower percentage. At reduced solvent rates on increased water concentration you may face loss of activity or loss of compounds. 100% DMSO best save the activity and compounds. WHile at test you will use minor concentration only as per the final concentration. For a concentrated extract you may have to go for 100% DMSO. And you may mention how much final concentration of DMSO is there in the assay broth (refer paper titiled final conc)

I am using DMSO for the same purpose and, as mentioned before, for the antimicrobial activity 100% may not be a problem.
There are different DMSO grades, HPLE, NMR, Laboratory grade, etc, which one you would prefer.
