Hi all! To monitor eukaryotic eggs or larvae cells in a coastal environment I was wondering if CARD-FISH or FISH would be a suitable tool? Or are there issues with permeabilization / treatment of those cells? Does anybody know about coral probes?

Any staining protocol has to be optimized. Usually, protocols include permeabilization steps (protease, collagenase,...). ISH-based protocols are conducted using high temperatures (up to 70°C) providing good probe penetration already. It depends on egg/larval shell composition whether or not additional treatments are necessary.
