What is the purpose of your assay? Are you interested in monosaccharides, polysaccharides or both? Do you want to measure total sugars or do you want assay each individual sugar. If the latter you will need to do some chromatography. Note that sugars (polar) will not be retained by the hydrophobic C-18 bonded phase. There are, however, a range of carbohydrate columns that can be used. I suggest you look at the application notes published by the different chromatography companies.
If you just want a total then perhaps a colourimetric assay would suffice (eg phenol-sulphuric acid). There are also a range of commercial kits (enzymic and colourimetric) available.
I want to measure both monosaccharides and polysaccharides in lignocellulosic biomass. and also want to determine each individualal type of sugar like mannose, arabinosse. gelucose and so on.
