I am trying to T cell suppression assay by separating myeloid derived suppressor cells from human PBMC. For this experiment, I used Jurkat cell line instead of autologous T cells. I want to know if this is possible. If not, please tell me why. I have just started immune cell experiments. So I have no basic knowledge.

I would recommend no to use Jurkat T cells as effectors. Jurkat can proliferate without stimulation and in suppression assay you would be looking how MDSCs affect TCR driven proliferation. It is better to isolate CD3 cells from the same individuals whose MDSCs you are using.
Jurkat cells will also proliferate very fast and would deprive MDSCs of the nutrients in coculture affecting their function.
