Can anyone tell me whether a pyrimidine derivative is possible from mannich product (beta keto amine) by condensation with urea with suitable catalyst? I have synthesized 2,4,6 tri substituted pyrimidine derivatives from chalcones, now looking for 1,2, 4 trisubstituted derivatives.
A 1,2,4-substituted pyrimidine would have a positive charge on the nitrogen. Do you actually want that, or do you mean a 1,2,4-substituted pyrimidone?

Thanks. What you said is correct. Please look at the possible mechanism I have worked out. It is not exactly a pyrimidine derivative, but I am specifically interested in these derivatives as a part of regidification hypothesis.

I believe that the amino group of the Mannich aminoketone would be more nucleophilic that the urea NH2. I've attached a variation of the mechanism which includes that, plus a paper which appears to confirm the idea.
