I am trying to determine the MIC for one of our strain that will be included in the GRAS dossier for the FDA. The strain we are testing is a Clostridia-class microorganism and we must do the experiment anaerobically, since the strain is strict anaerobe.
We don't wish to do the test in agar dilution, as it may complicate the tests. Can we do the test in micro dilution 96-wells setup for regulatory acceptance?

For C. difficile, the authors Hastey CJ et al. (PMID: 28188880) did not recommend the use of broth microdilution method as alternative to agar dilution for C. difficile antimicrobial susceptibility testing. The researchers analysed a huge set of isolates (n=920) against seven antimicrobials.
Agar dilution or E-test is the most common use methods for AST testing in Anaerobes in routine microbiological labs, so I think that it would be bette to overcome suggested complications to obtain the valid results.

Clostridia are difficult organisms to deal with, especially in terms of AST! I suggest E-test will provide the best alternative,as it uses clear cut results interpretation.
Anyway,anaerobiosis study needs replicative readings, with means taken and compared with standards!
