I'm using 10 mM Phosphate buffer to run HPLC, is it necessary to filter it prior ? or since degasser is there in the instrument can i go without filteration. Im standardizing right now but soon be moving with serum samples.
It must be ensured that all compounds that will pass through the column do not have a diameter greater than the space left by the polymer filling the column (normally specified by each column), otherwise it may clog. You can use HPLC grade solvents, but considering that you are going to use a buffered solution, I advise you to filter it before using it with a membrane with a specific pore. Filtration is not just for degassing and this is a general advice not having more information about your work.
Yes. Small, unseen, particles can block your frit and drive your back pressure very high and destroy your column (a high technology item). In addition there is no guarantee that 'other' stuff in your mobile phase or extractant will not preciptate (thus mobile phase preparation beforehand).
You will pay now, or you can pay later. You will save time now if you do not filter, but you will have to put up with check valve issue, pump seal, and the most important is the clogged column. It is your choice.
