Dear all,

I am using Is eFluor™ 670 eBioscience die (Invitrogen 65-0840-85) to stain NK cells. The datasheet indicates that it binds to any cellular protein containing primary amines and that it can be used for long term tracking of labeled cells. I am wondering if cells might loose the staining if cells die.

Basically, I do not achieve a uniform staining of all NK cells with Fluor™ 670 eBioscience, since some of them are much brighter than others. Also, I am wondering whether in some conditions I might not be visualising that many labelled NK cells in case cells lose the die upon cell death. Could anyone shed some light on this?

I am trying to track NK cells over 24h-48h (microscopy) and I would need a die giving a reliable staining for the cells so any help will be much appreciated.

Also, could Hoechst staining be an option? I know it perfectly stains all my NK cells, but does anyone know if Hoechst staining night affect NK cell functionality?
