I want to evaluate the growth of E. coli in a Varioskan plate reader equipment. I wonder if the equipment will be overloaded if I use the shaking function during several days. My OD reading will be between 0,1 and 0,7 OD600.
Equipment overuse is advised against. The term overuse is a derogatory word and nothing good can come out of it. Additionally, there is no duration of fermentation like "several days", as used by you. Accordingly, one cannot give you a definite answer.
We have been evaluating bacterial growth using our plate reader (Synergy H1) for up to 72 hours, under continuous shaking. So far we had no problems with the equipment.
You can evaluate E.coli growth by measuring OD at 600nm, also you can create dependence of CFU/ml of E.coli from OD at 600.
For calculation of CFU/ml you can use serial dilution method. You can measure OD at 600 of your E.coli suspension and then use serial decimal dilution method for calculation of CFU/ml. You can create curve for calculation CFU/ml from OD at 600 by calculation of At least 3 points of CFU/ml of E.coli suspensions with different OD.
I don't think that your equipment will have a problem but...One of the problems that you can face during long experiments in the microplate reader is evaporation. I think that as long as you are sure that evaporation is somehow controlled then the time for the fermentation should not be a problem... That being said I never run an experiment in the microplate for more than 36 h.
