Inhibition of histamine release from RBL-2H3 cells (mast cells) / Histamine rele, Problem in getting a reproducible result (histamine release assay)

I've been trying to optimize this histamine release for quite sometime and I face a problem in getting reproducible results for this assay.

The purpose of this study is to study the inhibition properties of a plant extract to inhibit the release of histamine (protein) from stimulated cells (RBL-2H3-mast cell line)

This assay is about, giving drug treatment to the RBL-2H3 cells (mast cells) before stimulating the cells to release histamine and to measure the rate of the inhibition of the histamine release from this cells after treatment with plant extract. Is there anyone who has done this assay before and managed to get reproducible results?

It has been quite a long time since you asked for advises... I just found your post as I would like to perform this assay on RBL-2H3 as well. I am aware of good response to IgE but very limited response to CP48/80 with these cells. Have you been able to use this cell line for such assay eventually?
