How should I induce diarrhoea in albino rats using diarrhoea-causing organisms? I am also looking for help on how to determine the infective dose i.e how many cfu/ml will be needed for the infection to be established.
Dear Dr. Busayo, Citrobacter rodentium has been long used as a surrogate model for EPEC/EHEC diarrhoea, in mice. I have no information about rats as animal models for C. rodentium, but I think it is worth looking at the literature. If you could take hold of a proper strain you would have no problems with finding an appropriate protocol for mice infection, there are many articles on the subject. I worked in induced diarrhoea in murine model (balb/c) previously. In my experiment I used the ETEC O78:h11 and it was very good choice. The infective dose will depend on the what signs you will be looking for. But forsure you will be able to get some ideas through scanning of some previous studies (try not to forget about the age and the weight of your animals and you can find the proper dose).
