I don't have any requirement in particle size, just need to grind samples as uniformly as I can, they have to be frozen (I can't dry them) and fibers tend to be a huge problem if I don't find a trick to get rid of them. Do you think that freezing and hardening with dry ice may help? Would you use liquid nitrogen + mortar instead?
PS I'm looking for water soluble carbohydrates by HPLC.
I had similar problems with plant tissues for DNA isolation. Liquid nitrogen + mortar + liitle bit of inert abrasive agent (i.e. carborundum or another ones) were very effective and helpful.
I agree for liquid nitrogen to quick freeze. For the grinding, mort/pestle works nice, but if you have a large number of samples, I would recomend automated bead grinders, for savinfg time and for grinding uniformity and reproducibility. Several brand exist, just use one with approriate bowl and bead sizes. Best regards.
