We quantitatively analyzed the number of muscle nuclei and focused on the segmental border muscle (SBM), which can be easily visualized due to its position. To mark the nuclei of all FCs and FCMs we used Dmef2 antibody. To specifically stain the nuclei of the SBM muscle we used the anti-Ladybird antibody. Ladybird is a muscle identity gene that is expressed in the SBM muscle.
