in order to the extraction of RNA from ovules how I can extract ovule from ovary without any change in expression profile? Does it need to be isolated in a special liquid or condition? are there any common protocols to extract ovules?

RNA isolation and library construction for transcriptome analysis :
Ovules were excised from 20–30 developing fruits at each developmental stage, in chilled (−20 °C) ethanol, under Leica M205 C stereo microscope in RNase free condition (Supplementary Figs S4 and S5). Total RNA was extracted by using Spectrum Plant Total RNA kit (Sigma Aldrich, St. Louis, MO, USA), following manufacture user’s protocol. DNase I (Sigma Aldrich St. Louis, MO, USA) treatment was given to remove genomic DNA contamination. The RNA integrity was established, using a 2100 Bioanalyzer (Agilent Technologies, Santa Clara, CA, USA). The equivalent quantity of total RNA extracted from the ovules of three biological replicates (plants) was pooled for transcriptome sequencing.
