I want to turn a regular cell culture incubator to a low oxygen incubator. Does anyone have experience with such a modification? I was thinking of splitting the CO2 tubing before the entrance and connecting it to Nitrogen and adding an O2 sensor inside. I would be happy to hear from someone with experience.

Sounds fun if you like constructing things, but if you really want low gas control in a standard incubator do not go remodeling your standard incubator...it is just fine. Instead, just get a box of the Petaka G3 cell culture cassette. It autoregulates oxygen and CO2 to the equivalent of 3-5% O2 and perfectly balanced CO2 and pH, using a standard incubator. In fact, with the Petaka G3 you just need a warming cabinet...no supplemental CO2 required. The low cost plastic disposable provides all the CO2 the culture medium needs to stay buffered. So leave the incubator intact and get Petaka (LOT for attached cells and FLAT for suspension cells). You will be glad you tried this new technology. I know I was the first time I used it.
