I want to improve the spore count of Bacillus amyloliquifaciens, which media is suitable? any one have media or strategy to improve the spore count upto 4 billion/ ml
Most isolates of Bacillus amyloliquefaciens only grow to a cell titer of ~2 x 10^8 cfu/ml at stationary phase in standard shake flasks. Even if sporulation is 100% efficient, you will need to concentrate the spores by centrifugation and suspension in a smaller amount of water or saline. The best liquid sporulation medium is probably strain dependent. But I have attached a couple of recipes that work well for me. One of them, 2XSG, is a complete medium, while the other, CSDM, is a defined medium. They work either as liquid or solid media. There are other options available, but I know these work.
