Zymogram analysis of β-glucosidase and β-galactosidase

·         The samples mixed with sample buffer(Non denaturing) and applied onto a polyacrylamide gel

(4% stacking ,10% resolving gels).

·         Electrophoresis was performed with 100 V at 4°C.

·         The gel was immersed in 3 mM 4-MUG and 4-MUGa in 0.1 M sodium 10 acetate (pH 5.5) for 10 min at room temperature to develop bands showing ß-glucosidase and ß-galactosidase activities, respectively.

·         The blue-fluorescent bands appear in a few minutes under UV.

·         4-methylumbelliferyl-ß-D-glucopyranoside (4- MUG) Sigma

·         4-methylumbelliferyl-ß-D-galactopyranoside (4-MUGa) Sigma
