I am trying to integrate FISH in our group, though I have some real problems with culturing the pancreatic INS-1E cells in ibidi 15ul-slide VI.
The majority of the cells seem to properly attach but they round up within the channels. They appear to grow well in the periphery of the channels just at the wells.
They don’t seem to be washed away. I thought it probably could be caused by routinely change of the medium that stressed the cells, or inefficient diffusion of nutrients etc. in the channel. But when using an ibidi open µ-Slide with 8 wells I experience the same pattern, where they grow good at the periphery, but are rounded in the middle of the wells.
I hope that you guys in here can clarify what the problem could be
