Are you referring to the rectangular block valve with handle for the mobile phase (valve that runs to waste or to the solvent container)? In either case, just remove this prep valve, unscrew it from the pump, disassemble and clean. If you have been running with any salts or buffers, then the tubing may be clogged with salts. Unscrew the front nut and pull out the 1/16" tubing. Replace the line with a large bore tube and flush the valve. If the valve is clogged, remove and clean the valve, then replace and flush before use.
Mr. Letter has suggested the best way. I would like to add the names of solvents that you can use for flashing the valve, i.e. Milli Q water and IPA; and also replaced thin tube can be placed in to a sonicator and using the sweep sonication option you can remove all clogged salts and other materials.
