I need to take IF images with DAPI on a confocal but I need to use a glycerol fixation. Has anyone done this method before, if so how was it done?

Glycerol is not a fixative... Do you have a paper/reference regarding this method?
Most common fixatives for IF include PFA and cold Methanol/Acetone.
In my lab we use glycerol to fix tissues/organs that need to be sliced! It’s prepared and the tissue fixed on a platter, then prepared glycerol is poured on the tissue, and left to dry, then cut to shape then sliced with the vibratone machine; I can give you the protocol for preparation
glycerol is added to the fixative to increase the density of the tissue once some glycerol embeds the tissue. You can see the tissue floating at the beginning (no glycerol embedding) and upon taking some glycerol it will decant to the bottom of the bottle/flask/container.
