Is your gold conjugate blocked with e.g. BSA? Is the sample serum or buffer?

or blocking with oligo(ethylene glycol) (thiol terminated)?

I used BSA (10%) after conjugation and it worked.

The false negative is from a human blood sample? You may wish to add heterophilic blockers to the assay: e.g. http://scantibodies.com/blockers/
