How to prepare the real samples for the determination of microcystin-LR in Protein phosphatase inhibition assay?

I am currently doing PP2A inhibition assay for determining Microcystin-LR. But in case of real environmental water, how to prepare the samples?

Microcystins in fresh water may be found free or inside cells. Usually, more than 90% are found as intracellular. Therefore, you need a procedure to break cells and recover microcystins. We optimise a specific procedure that works very well with PP2A inhibition assay.
