I see an option of "Endnote manuscipt" while uploading on journal website (attached screenshot with red highlighted). This option automatically extracts the information from single word file and saves time to fill each and every section while uploading. I tried looking for the that particular journal's Endnote manuscript format on web but couldn't find. There are some templates available in Endnote but those are very few and not of every journal. It would be very helpful if anyone will help me to work this out.
Endnote uses accollades to put citations in your text. During manuscript processing on a journal's web page, it is common that a pdf is constructed from the files you upload.
It is my guess that the option you highlighted passes instructions to the webpage to interpret the citations in your manuscript correctly and request the original endnote database as well.
Templates or "output styles" in Endnote are something completely different. You access those from the menu: Edit - Output Styles - Open Style Manager. You'll notice that the list for different journals is long. Very long. Place a tickmark before any of the journal styles you want to format your references for.

Thank you for your reply. I am aware of the output styles in Endnote for citing references in the word file. However there is this option in Tools - Manuscript Templates - different journals pre-formatted word files, that's the one I am talking about where options are not much. Further while uploading in the highlighted option (image), when you upload the word file, it extracts title, abstract, text, references etc when the manuscript is written in the pre-formatted word template. I am assuming that there must be more word templates for every journal which is not mentioned anywhere on the corresponding journal's website. This saves time but I am wondering that without giving any instructions why even this option is mentioned.
The feature is rather new. With 193 references in the latest version of EndNote, it might expand in the not too far future. I don't get the thoughts behind a journal's choice of not providing a suitable template. I would go Darwin on them and find another journal that does. :)
