i'm working on project to optimizing dye degrading bacteria with respect to its pH, temperature and dye concentration. in order to optimize the pH and temperature, what is the suitable concentration of dye that i must add in the broth media? or should i conduct study on optimizing the dye concentration first, then the result will be used in optimizing the pH and temperature?

Hi,
First find out effect of varied initial dye concentrations (100 mg/l - 600mg/l) on decolorization. Then use this optimum dye concentration for checking effect of Temperature (20 - 40 degree centigrade) and pH (4 to 9) on dye degradation. You can even check effect of static and shaking conditions as well as carbon and nitrogen sources in the media on decolorization.
