As I understand if you use the mean and SD for describing your data, this means it is normally distributed and parametric. If so, the test for variance should be simple T test.

It is preferable to mosaic the satellite images by "mosaic to new raster" from Arc Toolbox, Data management Tools, Raster, Raster Dataset, Mosaic to New Raster.. before performing the classification and change detection.
