I'm trying to develop method for Resorcinol Detection on LC-MS/MS but it seems it is nasty and sticks on the column just like phenol.
The machine has an Alltima C18 Column, 250×4.6 mm, particle size 5 μm, from Alltech Associates Inc. (Deerfield, IL, U.S.A.) to be used at 40°C for the chromatographic separation.
I am mainly looking for what mobile phases to use and in what gradients? And which polarity for MS?
I already tried several tests with (A) H2O and (B) Methanol+Acetonitrile mobile phases but i could optimize a reliable peak.
And also tried (A) H2O and (B) Acetonitrile and no clear peaks in LC. Worse, when I scan MS, i get resorcinol all the way from 3 min to 20min continuously showing up at like 10^8 cps although injection volume is 20uL.
Any comment/feedback would be highly appreciated!
