I'm trying to develop method for Resorcinol Detection on LC-MS/MS but it seems it is nasty and sticks on the column just like phenol.
The machine has an Alltima C18 Column, 250×4.6 mm, particle size 5 μm, from Alltech Associates Inc. (Deerfield, IL, U.S.A.) to be used at 40°C for the chromatographic separation.
I am mainly looking for what mobile phases to use and in what gradients? And which polarity for MS?
I already tried several tests with (A) H2O and (B) Methanol+Acetonitrile mobile phases but i could optimize a reliable peak.
And also tried (A) H2O and (B) Acetonitrile and no clear peaks in LC. Worse, when I scan MS, i get resorcinol all the way from 3 min to 20min continuously showing up at like 10^8 cps although injection volume is 20uL.
Any comment/feedback would be highly appreciated!
You could try detecting this compound in both positive and negative polarity. I would test ammonium formate/ammonium acetate with formic acid as additives for your mobile phases and look for M+H, M+NH4 or M-H ions. Such a small compound would show little retention in RP-LC. An acidic eluent can help reduce excessive interactions with active sites in your column. If possible, test another column that could provide you alternate interactions with your analyte, e.g. C18 Amide, aqueous C18, fluorinated phases.
