You need to look and read carefully if it is 50 ml or µl.

When a protocol say use a toothpick to pick cells, it refers to the amount of cells which can be transferred using the tip of a toothpick.

If you are still unsure, just ask any experienced person in you lab.

I guess your protocol is from a Nature paper (see below), so '50 mL' in your question should be a typo. You should change it.

"Scrape a 50 μl volume of yeast from the YPAD plate using a sterile inoculating loop or sterile toothpick and suspend the cells in 1.0 ml of sterile..." (https://www.nature.com/articles/nprot.2007.14?foxtrotcallback=true)

Just take a PCR tube, and add 50 uL water in there to get an idea of the amount of 50 uL water. Then, take similar amount of the scraped cells from the plate. Then, you should have ~50 uL of the cells.
