Few pesticides like quinolphos are very less soluble in water(solubility-17.8 mg/L) but are highly soluble in organic solvents like hexane( solubility- 250000 mg/L). My aptamer library is dissolved in TE buffer. How should I prepare stock solution of pesticide to perform SELEX. If pesticide solution is prepared in hexane(organic solvent), is their possibility that immiscibility of solvents ( TE buffer and hexane) will affect various steps of SELEX? Maximum possible stock solution of quinalphos in water is 0.0596 mM and in hexane is 838mM.
Have no idea about compatibility of hexane with TE buffer. But, use of acetone to prepare stock solution of quinalphos may be explored. Quinalphos solubility in acetone is >1000 mg/L and should be compatible with TE buffer.
Firstly take Quinalphos and Dissolve with some amount of Acetone and make up with Hexane I think 1000 mg/L and should be compatible with TE buffer.
