Sorry if any of you found this question a bit silly. But, May I know, if I am doing an investigation on medicinal plant and would want to know its antioxidant activity using DPPH scavenging activity (Note:the extract was prepared by dissolving 2g of dried plant in 20 ml solvent):
1) Is my crude extract was considered to have a concentration of 0.1g/ml (since in many DPPH protocols, need to have a serial concentration of samples tested)?
2) So, if it is considered as 0.1g/ml, then, my dilution should be from this point right?
3) Usually, what is the blank, the control used in DPPH scavenging activity? As I have seen some differences about the blank and the control, as well as equation used in literatures.

Yes your concentration is 0.1g / ml. You should use this solution to make the dilutions. The control is usually the solvent you use for the extraction.

1. Yes, your serial sample concentration would be less than 0.1g/ml.
2. Yes, your dilution sample would start from this point.
3. We should make control the same as no.1 (series of concentration sample) without adding DPPH.
Be carful that DPPH method is very affected by pH of extract which gives vague values. Therefore, better to adjust pH of the extract at 7.

Mainly dilution depends on extract antioxidant activity. It will vary sample to sample.
You may start 0.1g/mL, if no absorption change than you more dilute and try. In my practical experience ....May be need more dilution
