Il have a same answer. But u can also use ethanol instead of butanol for preparing solution. I prepare without acetic acid than also the charring pattern was good.

I've used ninhydrin solution to stain amino-phospholipids in TLC plates. The staining solution was prepared by weighing 0.2gr in 100mL of butanol saturated with water (vortex butanol in water and collect the lower phase). Hope this helps,

Dissolve 0.2g of ninhydrin in 100ml ethanol. Mix thoroughly.

I have also same answer.

Dissolve 0.2g of ninhydrin in 99.5ml ethanol and 0.5 ml acetic acid. 

1.5 grams Ninhydrin

100 ml Butanol

3 ml Acetic acid

0.2 grams Ninhydrin in 100 ml Acetone works equally well.

8 % (w/v) Ninhydrin solution- 8 g of ninhydrin dissolved in 100ml acetone and keep on cold condition for future use.

1.5 gr of Ninhydrin

100 mL Butanol

3 mL acetic acid

What is the role of acetic acid? Is it necessary to add to the solution?

Optimum/maximum colour development with ninhydrin reagent is in the pH range of 4 to 6; that is why acetic acid is added.

Dissolve 4 g of ninhydrin in 100 ml ethanol and mix well without acetic acid

Solution of 5 g ninhydrin in 95% EtOH dissolve well and work well.

It works better if you prepare a 0.2%(w/v). Add 0.2 g of ninhydrin in 100 ml of ethonol. You can dissolve thoroughly using ultrasound.
