I want to functionalize gold nanoparticles with apolymer for drug loading, and I also want to fuctionalize it with tumour directing ligands. what should be the steps, should i first functionalize with the polymer , than with tumour directing ligands, or i can do it in a single step.
Hi Aiman, Au particles are very refractive to modification.
The easiest way to modify them is to first use mono -SH compounds with either -NH2 or COOH end groups that can be further modified.
The simplest way would be to purchase commercial citrate-stabilized colloidal gold nanoparticles (Ted Pella sells them in various sizes: 2, 5, 10 nm...) and then simply mix them with thiolated ligands of your choice. If your polymer and your tumor-directing ligand both have a thiol (-SH) group, you can do this in one step. However, a major challenge is to determine the resulting stoichiometry of the two molecules on the gold surface, since adding them in a 1:1 ratio in the reaction mixture will not ensure 1:1 loading on the gold surface. You can characterize the surface loading of your particles by dissolving them with NaCN or KCN, then elute the mixture through a gel filtration column (Sephadex works well). Then, quantify the amount of polymer and tumor-targeting ligand in the eluent by HPLC, UV/Vis, etc.
