PCR amplification: If the water mites are too small or difficult to isolate, PCR amplification can be used to directly amplify DNA from the insect's tissue, including any water mite DNA that may be present. This method can be effective for detecting low levels of mite DNA in insect samples.

When it comes to separating water mites from insects while preserving the mites' integrity, a technique called "surface tension separation" can be employed. This method takes advantage of the differences in surface tension between water mites and insects.

Freeze the mite-infested mosquito for 30 minutes or more at -20 deg C or, better still, -70 deg C.

Remove mites with fine forceps under 40x image magnification.
