I want to specifically target Flavonoids present in my chloroform plant extract. Is there any specific solvent or method through which I could focus on flavonoids mainly?
Actually two different important points are presented in your question; first chloroform is very known to extract lipids or alkaloids so it is strange that you search flavonoids through extraction using this solvent; secondly; you should get Silyl Derivatives flavonoids to be able to use GC-MS.
For my opinion; I suggest changing the exctraction solvent than use HPLC-MS or UPLC-MS; both are the best in flavonoid identification.
Khedidja Benarous Respected mam thank you so much for your kind response. My chloroform extract showed maximum biological activity whilst other(methanol, ethanol, water) couldn’t.

I suggest 10 or 20 mg as enough concentration for LC-MS; and your biological activity of the chloroform extract surely depends to alkaoids; you should search if your plant is rich with alkaloids or not; if it is than you have a very good result just continue further analyses to publish your work
In my humble opinion, it is better to HPLC and LC/MS. The GC/MS does not really favorable results for flavonoids and phenolic compounds. It is essential to prepare the respective standards solutions and draw the calibration curve before starting the test. It is strongly recommended that it was prepared some prototypes and the result obtained were compared with principal samples/analytes.
