I am giving infection to the plant with rhizogenes, where i am seeing leaching at the same time thread like structures tooo. I am in bit of confusion with HRs and other roots.
Use rolB or any such Agrobacterium specific PCR primers, set a PCR for the different roots and confirm the same. Hairy root would have the agro-specific plasmid DNA fragments that would amplify. If I am right, then upon microscopic observation, hairy roots would not have root hairs!
Thank You for your answer. If we run a DNA with a primer set, we have no problem to identify. But My problem is sir, to identify the hairy roots with naked eye for further co-cultivation.
I suggest to cultivate your plants in the way that wild type roots remain in the agar-based media, and the place of A. rhizogenes infection remains outside (try to keep a good humidity in the plate to promote hairy root formation), the resulting hairy roots are going to be easily identifiable by the fact that they are not going to try to reach the culture agar-based media and keep their growth (hairy roots are not affected by the gravitropism).
