Hi. There are many strategies to solubilize proteins. You can use inclusion bodies solubilizing buffer (composition available in Sambrook et al) or 1% SDS or Urea (around 4 - 8M) or even a combination of these and treat your pellet. I am not sure which is your expression system but if it is a bacterial expression system then you can even reduce the incubation temperature around 16-25 degree Celsius. If you use IPTG for induction then you can reduce the concentration of that too and look for protein solubilization. I hope this would help you. Good luck.
in brief; use a solubility enhancer tag(MBP, Trx, etc), test overnight time in 12-18 C, use 0.5-0.4 mM IPTG for induction and use enriched broth such as terrific broth or 2xYT.
hI...Shubhashish Chakraborty..Thanks for your explanation,im working with PET22b expression system..my protein is being collected in the pellet, seems like there is no band size in the SDS PAGE gel for supernatant sample..
