Seasonal greetings
I want to estimate some important soil enzymes such as α, β glucosidase and α, β- galactosidase from soil. In brief, I have applied some biofertilizers in the soil. I want to observe the response of enzymatic activity.
Please let me know if standard and simple procedure is available. I have the protocol described by Eivazi and Tabatabai (1988). But, this procedure is time consuming and costly as the chemicals are very costly.

- The principal of the method described for assay of alpha, beta glucosidase activity are similar to those of phosphodiesterase activity. The method is based on colourimetric determination of the p-nitrophenol released by glucosidase. when soil is incubated with buffer pH 6.0 PNG solution and toluene. the incubation is carried out at 370 C for one hour. the p-nitrophenol released is extracted by filtration after addiction of 0.5M CaCl2 and 0.1M THAM buffer at pH 12.
Assay method
- Reagents- 1. toluene, modified universal buffer (MUB) pH 6.0, Calcium chloride (0.5M), 0.025 M PNG solution- dissolve 0.377g of PNG ( para nitrophenyl -beta-D- glucoside) in about 40ml of MUB pH 6.0 and agjust the volume to 50ml with the same buffer. Stir the solution in a refrigerator and tris (hydroxymethyl) aminomethance (THAM) buffers (0.1M), pH 12 and 10.
