The target protein was downregulated after siRNA transfection for 48h. I want to do CCK8 experiment or other functional studies. I design: 1 Mock(only cells), 2 lip( no siRNA), 3 NC ( RNAi negative control+lip ) , 4 siRNA. If I directly do the CCK8 after siRNA transfection. But I donot kown for 48h or other time？And lip have Cytotoxicity，if i should take the lip as the control and overcome cytotoxic effects?
I also consider another option. Counting the transfected cells again and then culturing cells to the new plate. But i donnot know the time: 1 the transection time, 2 to the new plate for CCK8 time??
