Is there is any thorough procedure for this analysis? I would like to know about the procedure to conduct Hypsometric analysis of a watershed, data required for it etc.

You can use Microdem software which is freely downloaded from the web.
Software requires only a clipped DEM (GeoTiff) of the catchment.
When you add the DEM chose "elevation histograms" from the "analyze" menu.
