I am trying to dissolve 2.3g/l of sodium hyaluronate in phosphate buffer saline. However, it is taking me two days to dissolve. I am wondering if somebody knows a way to dissolve it faster. I can't apply heat for more than 37, which is the body temperature, right?
Solubilization speed varies with the MW of the hyaluronate, lower MW have faster dissolution.
Sonication helps solubilizing on some cases, I don't know if it would help for this though
Yes my molecular weight is 1000000 Dalton. Unfortunately, We don't have Sonication in our lap
unfortunaly it need lot of time to solubilize HA.
with a powerful Falcon tube shaker i needed 24 h hours

Yes...i think that the only way is to degrade it termically in an oven. max 60C if i can recall to have a slowly degradation
but it depend on what you need to do :D
Probably I state the obvious, are you adding the HYal power to the solution? If you do the opposite the soulibilization should be more difficult.
you can also try to lyophilizate it. The resulting Hyal (the aspect is like cotton ball) will be easily solubilized.
