I had previously frozen mouse limb buds in liquid nitrogen and am now attempting to dissociate the cells for CUT&Tag. However, attempts with multiple collagenases and accutase result in very low cell yield (and most of the cells are not intact).

Does anyone have any recommendations for other methods of dissociation? For reasons specific to the project, the tissue must be snap-frozen before processing.
