Wash the cells with cold PBS (without Ca2+/Mg2+). Then treat cells with cold PBS-EDTA (5mM) for 10 mins. This works quite well.

I am using Accutase and incubate at 37˚C for 10 min.

It can help you get about 50% of the cell.

You can also use cold PBS to wash and use a cell scraper to get the rest of them.

Fan Zhang wouldn't scraping the cells damage the receptor present on it.?

From my experience, it works just fine. They were able to be stained by CD86 and CD206.
