Please help me, how to convert glucose reading from analox instrument to Glycemic index..
About 300ul of rice extract in 150ul of PBS and 20ul of the alpha glucosidase enzyme was added and glucose response was checked in an Analox instrument from 0 to 120 mins at 0, 30, 60 , 90 and 120 min time intervals.Each injection volume of sample was 10ul.
Gucose response for example:
0.4, 9.1,18.5, 26.8, 34.3 Glucose(mmol/L) at 0, 30, 60 , 90, and 120 min time intervals respectively.
Area under curve for this time vs glucose is =2153 mmol/L/m2
So how to convert this data into glycemic index?

The Glycemic Index is a numerical Index that ranks carbohydrates based on their rate of glycemic response (i.e. their conversion to glucose within the human body). Glycemic Index uses a scale of 0 to 100, with higher values given to foods that cause the most rapid rise in blood sugar. Pure glucose serves as a reference point, and is given a Glycemic Index (GI) of 100.
Glycemic Index values are determined experimentally by feeding human test subjects a fixed portion of the food (after an overnight fast), and subsequently extracting and measuring samples of their blood at specific intervals of time.
GL = GI/100 x Net Carbs (Net Carbs are equal to the Total Carbohydrates minus Dietary Fiber)Therefore, you can control your glycemic response by consuming low-GI foods and/or by restricting your intake of carbohydrates.
1. First you have to determine the weight of "digestible carbohydrates" content in your rice sample.
2. Then you have you have to take a similar weight of pure glucose (if purity is not 100%; adjust the quantity by calculations) and measure absorbance values in the same time intervals as you have done earlier.
3. The calculate;
(Incremental area under curve for rice / IAUC for Glucose)*100
That will give you the GI.
But remember; that you have to be very precise and accurate with your digestible carbohydrate calculations; and then glucose portion size confirmation.
