I recently got a bacterial strain with a ropy phenotype. When I attempted to collect the cell pellets from broth culture using a centrifuge, I found the cell pellets were difficult to precipitate because of the viscous property of the culture. Do you have any advice? thank you very much.
Hi: The culture maybe to old and that allowed the bacteria produce a ropy product in media, you should grow it may be less then 48h
Or, the media that you prepare is have some ingredient that make the culture ropy, you should use another media to grow the bacteria.
1st - what kind of bacteria are you growing?
2nd - which media are you using? Did you prepared ti by your self or you got it from a company?
i agree with Mutiullah, use small/ dilute inoculum. I would like to know the appearance of bacterial growth in broth, was it uniformly suspended, at bottom or slimy etc.
It was uniformly suspened in the broth, and the colonies on agar plate were also ropy. The bacteria were Bifidobacteria, and the culture media was MRS.
try to centrifuged at 8590g for 1 hour at 4°C, and then collect the precipitate or dissolve the precipitate in buffer solution if you do not need the bacteria in precipitate form.
